Have you tried rebooting your computer and then reinstalling from http://altitudegame.com/download.html ? Have you tried updating your video drivers directly from nvidia.com ?
Please zip up all the files in ~Altitude/log and email them to support@nimblygames.com

Drivers are up to date. I haven't had trouble with it before, pretty sure it's just some really wonky corrupted file.
Drivers are up-to-date, and I tried installing the game from that link into another directory, didn't work.

Did you try rebooting your computer? This file shouldn't be locked, but it is:
C:\Users\[YOUR_USERNAME]\AppData\Local\Temp\altitude-game-run-lock.tmp
C:\Users\[YOUR_USERNAME]\AppData\Local\Temp\altitude-game-run-lock.tmp

For future reference (I hope you don't mind for the link being here admins), this is a handy program I use called Unlocker - It's freeware and it detects all the processes locking a file when you right click on it and select "unlocker". You then have the option of killing the process or "unlocking" it - I'd recommend unlocking before killing processes.
